Surface tension, gravity. Single cells aggregate to create spheroid.
Limitations: Laborious, requires humidity, produces minimal droplets.
U or V shaped ULA plate
Self-assembled tumoroids cultivated as free-floating.
Limitations: Drug distribution hindered by high compaction in spheroid.
Matrix embedding hydrogels or polymers
Cells are seeded at 4C in synthetic medium and stiffen at 37C.
Limitations: Batch to batch irregularities. Needs enzymatic recovery.
